Standard operating procedures for biosafety cabinet

Objective: to standardize the operation and maintenance of the biosafety cabinet, ensure the normal operation of the equipment, and ensure the safety of the operators.
Scope: biosafety cabinet used in BSL-2 laboratory, ESCO ac2-4s1 and ac2-3s1.
Responsibilities: each operator is responsible for the normal operation and maintenance of the equipment during the experiment.


Standard operating procedures
Matters needing attention

1. The safety cabinet shall be placed in the primary purification room below 100000 level. Turn on the UV light before the first operation every day, irradiate for 30min, and then move all the items required for this operation into the safety cabinet to avoid frequent passing through the air curtain and damaging the air flow; and wipe the surface of the items with 75% alcohol for disinfection before moving in, so as to remove the pollution.
2. Turn on the fan for 10min, and conduct the experiment after the air in the cabinet is purified and the air flow is stable. Slowly extend your arms into the safety cabinet, and keep still for at least 2min to stabilize the air flow in the cabinet before operation.
3. Do not put anything irrelevant to this experiment in the biosafety cabinet. The articles needed for the experiment should be as simple as possible to avoid covering the grating of the cabinet with articles and minimize the impact on the air flow. The articles shall be placed as far back as possible, and the airway opening shall not be blocked, so as not to interfere with the normal flow of air flow.
4. Avoid cross contamination during operation. In order to prevent possible splashes, 75% alcohol spray bottle and paper towel should be prepared to wipe as soon as possible after splashing.
5. During the experimental operation, the glass window shall not be opened completely, and the operator's face shall be on the working window. The operation in the cabinet shall be gentle and soothing to prevent air flow in the cabinet.
6. The safety cabinet shall be inspected and maintained regularly to ensure its normal operation. Once the safety cabinet is found to work abnormally during the work, it shall stop working immediately, take corresponding measures, and notify the instrument administrator.
7. After the experiment, close the glass window, keep the fan running for 10min, and turn on the ultraviolet lamp to irradiate for 30min.
8. The safety cabinet shall be cleaned and disinfected regularly, and the worktop and the external surface of the cabinet can be wiped with 75% alcohol; after each inspection, it shall be fully disinfected.
9. The articles used in the cabinet shall be disinfected by spraying 75% ethanol on the surface and then taken out, so as to prevent the potential pathogen residues from polluting the environment and causing biological hazards.

usage method:
1. The safety cabinet is controlled by LED liquid crystal panel. The function keys included in the control panel are: power on key, sterilization light key, wind stop key and lighting key.
2. After connecting the 220 V AC power supply, press the power switch shown on the control panel to turn on; when sterilization is required, press the sterilization lamp key; and turn on the fan, as the case may be; when lighting is normally operated, just press the lighting key.
Equipment maintenance
1. After each inspection operation, use 75% alcohol to thoroughly clean the surface of the working area, side wall, back wall and window inside the safety cabinet. Do not use fungicides containing chlorine as it may damage the stainless steel structure of the safety cabinet. At the same time, clean the surface of UV lamp and power output port.
2. Clean and maintain the stainless steel surface every two weeks if it is not tested for a long time. Clean the stainless steel surface regularly according to the above plan to keep the surface smooth and beautiful.
3. Wipe the external surface of the safety cabinet with wet cloth every month, especially the front and upper part of the safety cabinet, and clean the accumulated dust. And check the reasonable use of all maintenance accessories.
4. According to the actual situation, check any physical abnormality or failure of the safety cabinet every quarter or half a year, and report for repair in time if there is any abnormality; remove and clean the primary effect filter to prevent dust accumulation, which will lead to insufficient air intake and reduce the cleaning effect.
5. Every year, the qualified certification technician shall conduct performance certification on the safety cabinet, and replace the UV lamp according to the service life effect of the UV lamp; when the air filter with normal adjustment or cleaning primary effect still fails to reach the ideal cross-sectional wind speed, the working voltage of the fan shall be adjusted (rotate the knob) to achieve the ideal uniform wind speed (adjust the knob from the left To the right, adjust slowly from low speed to high speed, and the new workbench should not be adjusted to the maximum wind speed).
6. After 18 months of use, when the working voltage of the fan is adjusted to the highest point and the ideal wind speed is still not reached, it means that the high-efficiency air filter has too much dust (the filter hole on the filter material has been basically blocked, and it should be updated in time), and the service life of the general high-efficiency air filter is 18 months; when replacing the high-efficiency air filter, pay attention to the model, specification and size (configured by the original manufacturer )According to the arrow wind direction device, and pay attention to the surrounding sealing of the filter, absolutely no leakage.
